Abstract

In this study, the effect of carvacrol, thymol, zinc and selenium on the metabolism of ethanol in the liver was investigated as theoretically The molecules of ethanol metabolism in gas, liquid and blood phase, structural parameters along with physical and thermodynamic properties of the studied molecules were calculated using Hartree-Fock(HF), Density Function (DFT/B3LYP) methods with hybrid approach and 6-31G (d,p)Basic set.These active ingredients (carvacrol, thymol, zinc and selenium), in which step of ethanol metabolism effective is illustrated by calculating the values of the free energy. We also compared with the related experimental studies. HOMO, LUMO, dipole moment values and hydrogen bond formation of carvacrol, thymol with ethanol, acetaldyde and acetate in the ethanol metabolism were measured. We also compared the related experimental studies. Consequently, ethanol metabolism in the liver that occurs in microsome, cytosol and peroxisome carvacrol is more effective than thymol. In addition, as the caculted values, zinc is exothermic, selenium that is not a direct effect in the metabolism of ethanol is endothermic, but it is to be effective presence of the structure of glutathione peroxidase in collecting free radicals can be said to be effective. To prevent loss of time and material, antioxidant compounds with known activity or event in a new unknown antioxidants by using computational methods give direction to the experimental studies can be determined.

Bu çalışmada karvakrol, timol, çinko ve selenyum?un karaciğerde etil alkol metabolizmasına etkisi, teorik olarak incelenmiştir.Metabolizmada yer alan moleküllerin gaz, sıvı ve kan fazında, fiziksel ve termodinamik özellikleri ile yapısal parametreleri Hartree-Fock (HF) metodu ve Yoğunluk Fonksiyonu Teorisi (DFT/B3LYP) hibrit yaklaşımı ile 6-31G(d,p) Temel Seti kullanılarak hesaplanmıştır. Bu etken maddelerin (karvakrol, timol, çinko ve selenyum), etanol metabolizmasının hangi basamağında etkili olduğu serbest enerji değerleri hesaplanarak gösterilmeye çalışılmıştır. HOMO, LUMO, dipol moment değerlerine ve karvakrol ve timol?ün etanol metabolizmasında yer alan etanol, asetaldehit, asetat ile hidrojen bağı oluşumlarına bakıldı. Ayrıca bununla ilgili yapılan deneysel çalışmalarla, elde ettiğimiz teorik sonuçlar kıyaslanmıştır. Sonuç olarak karaciğerin mikrozom sitozol ve peroksizomunda gerçekleşen etanol metabolizmasında; karvakrol?ün timol den daha etkin olduğu tespit edilmiştir. Ayrıca hesaplanan değerlerden çinkonun ekzotermik, selenyumun ise endotermik olarak olarak etkili olduğu yani selenyum etanol metabolizmasında doğrudan etkili olmadığı ancak glutatyon peroksidazın yapısında bulunması ve serbest radikalleri toplayarak etkili olduğu söylenebilir. Etkinliği bilinen bileşikler yardımıyla yeni bulunan veya etkinliği bilinmeyenler için zaman ve madde kaybını önlemek amacıyla, hesaplamalı yöntemler kullanılarak deneysel çalışmalara yol gösterebileceği belirlenmiştir.
