Abstract

The total RNAs (2.50 ?l total RNA +7.5 ?l RT mix and 2 ?l cDNA +23 ?l PCR mix), obtainedfrom dormant tubers belonging to the cvs Agria, Granola and Morfano which were infected withPLRV, PVS, PVX and PVY, were tested with uniplex RT-PCR and the bands, were taken fromeach of these three cvs intensive enough to be photograph. In the multiplex RT-PCR workingcDNA was synthesized with 2.5, 3.5 ve 5 ?l for all uses of the total RNA, but the best result wasfound when 5 ?l RNA were synthesizes. At the PCR stage when the different quantities of thecDNA and PCR mixture (2 ?l cDNA +23 ?l PCR mix, 5 ?l cDNA +25 ?l PCR mix with 5 ?lcDNA 20 ?l PCR mix) were compared the best result was obtained from the mixture 5 ?l cDNA+25 ?l PCR. At the same time the bands, which were intensive enough to be displayed in UVfor PLRV, PVS and PVX were only obtained if 25 ?l of PCR product was loaded to gel and nospesific band determining to PVY was obtained from any practice. As a result if m-RT-PCRwill be used for defining the viruses PLRV, PVS, PVX and PVY from dormant tubers, in orderto have beter display in UV, at the cDNA stage it must be synthesized by using 0.4 ng/?l oligodT primer, 20 ?g/?l spesific primer (for PLRV), 5 ?l total RNA and at the PCR stage the least3.5 ?l of cDNA must be used, the product of PCR must be loaded to gel 25 ?l, %1.5 gel mustbe prepared and it must be spilled thinly. In addition to these it is suggested that if it is possible,PVY must be dependent on PCR differently by using the same cDNA.KeyWords: VIRUS, PCR, m-RT-PCR, POTATO, CERTIFICAT.

PLRV, PVS, PVX ve PVY virüslerinin hepsi ile enfekteli olduğu belirlenen Agria, Granola veMorfano çeşitlerine ait dormant yumrulardan elde edilen total RNA'lar (2.50 ?l total RNA +7.5?l RT karışım ve 2 ?l cDNA +23 ?l PCR mix) uniplex RT-PCR ile test edildiğinde her üççeşitten de bütün virüsler için fotoğraflanabilecek yoğunlukta bantlar elde edilmiştir. MultiplexRT-PCR çalışmasında cDNA total RNA'nın 2.5, 3.5 ve 5 ?l'lik miktarları kullanılaraksentezlendiğinde en iyi sonuç 5 ?l total RNA uygulamasından; PCR aşamasında ise cDNA vePCR karışımın farklı miktarları (2 ?l cDNA +23 ?l PCR karışım, 5 ?l cDNA +25 ?l PCRkarışım ile 5 ?l cDNA 20 ?l PCR karışım) mukayese edildiğinde en iyi sonuç 5 ?l cDNA +25?l PCR karışımdan elde edilmiştir. Bununla birlikte PVX, PLRV ve PVS için UV degörüntülenebilecek yoğunluktaki bantlar ancak PCR ürünlerinin 25 ?l olarak jeleyüklenmesinden elde edilirken hiçbir uygulamadan PVY'ne spesifik bant elde edilememiştir.Sonuç olarak; multiplex RT-PCR dormant yumrulardan PVX, PVS, PLRV ve PVY'nünbelirlenmesi amacıyla kullanılacaksa cDNA aşamasında 0.4 ng/?l oligo dT pirimer, 20 ?g/?lspesifik pirimer (PLRV için) ve 5 ?l total RNA kullanılarak sentezlenmesinin, PCR aşamasındaise cDNA'nın en az 3.5 ?l kullanılması, PCR ürününün jele 25 ?l yüklenilmesi, jelin %1.5'likhazırlanıp mümkün olduğu kadar ince dökülmesinin UV'de daha iyi bir görüntü elde edilmesiaçısından gerekli olduğu saptanmıştır. Bunlara ilaveten PVY'nin mümkünse aynı cDNAkullanılarak ayrı olarak PCR'a tabi tutulması önerilmektedir.Anahtar Kelimeler: VİRÜS, PCR, m-RT-PCR, PATATES, SERTİFİKASYON.
