MALAT1 (metastasis-linked lung adenocarcinoma transcript 1) is defined as an oncogene long coding RNA having length of 8000 bp that promotes tumorigensis. MALAT1 has role in various biological processes. It participates in the post transcriptional processing of mRNA. MALAT1 has been found to regulate B-MYB gene, an oncogenic transcrition factor. In this thesis, we aim to investigate the possible relationship between the lncRNA MALAT1 and the B-MYB gene. MALAT1 inhibits several miRNAs and regulates gene expression of genes in lung, breast and bone cancer cell lines and therefore its effect on proliferation and apoptosis is being investigated.In this study, three different cancer cell lines were studied. They were lung cancer cell line (A549) andits control cell line BEAS-2B, breast cancer cell line(MCF-7) and its control cell line hTERT-HME1, bone cancer cell line (U2OS) and its control cell line HUVEC. miR-503, miR150 and miR-15a were used to inhibit MALAT1.In the experiment, the cells were cultured and incubated for 48 hours after the transfection of respective miRNAs. For gene expression analysis, total RNA was isolated and transformed to cDNA.The gene expression was determined by RT-PCR.Biochemical parameters like glutathione peroxidase (GPx), catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO), Myelin peroxidase and Prolidase activities were also studied in the cell suspension. Flow cytometry was used to determineapoptosis using the kit containing Annexin V for and 7AAD and cell cycle levels in cell lines using the kit BD Cycletest ™ Plus DNA Reagents. Gene expression was analyzed. MALAT1 and B-MYB expressed at different levels in different cancer types. As a result of biochemical studies, statistically significant differences were found between respective cancer and control cell lines in terms of all parameters (p<0.05).When we analysed cell proliferation and apoptosis, there was an increase in the number of cells undergoing apoptosis while there was decrease in proliferation in lung cancer, while in breast cancer, there was no change in apoptosis but there was an increased proliferation. In bone cancer, there was an increase in the number of cells undergoing apoptosis as well as there was increased proliferation.As a result, MALAT1 plays different roles in different cancers.While MALAT1 can be used as a powerful biomarker in lung cancer, it can also be used as a biomarker for proliferation in breast cancer and for apoptosis in bone cancer.

MALAT1 çeşitli biyolojik süreçlerde görev alır. Transkripsiyonel sonrası mRNA'yı işler. Kanserde aşırı ifade olur. Onkojenik transkipsiyon faktörü olan B-MYB geni düzenler. Bu tez çalışmasında ki amacımız, MALAT1 lnRNA'sı ve B-MYB beni arasındaki ilişkiyi incelemektir. MALAT1'i spesifik mikroRNA'larla inhibe ederek, akciğer, meme ve kemik kanseri hücre hatlarında gen ifade düzeyi, proliferasyona, apoptozise olan etkisinin olup olmadığını araştırmaktır. Çalışmada üç farklı kanser hücre hatları çalışıldı. Meme kanseri hücre hattı MCF-7 ve kontrol hücresi hTERT-HME1, akciğer kanser hücre hattı A549 ve kontrol hücresi BEAS-2B ve kemik kanseri hücre hattı U2OS ve kontrol hücresi HUVEC hücre hatları çalışıldı. MALAT1'i inhibe etmek için miR-503, miR150, miR-15a mikroRNA'ları kullanıldı. Deneyde hücreler ekilip mikroRNA transfer edildikten sonra 48 saat inkübe edilip hücreler kaldırılmıştır. Gen ifadesi için total RNA eldesi edilip cDNA'ya çevrilmiştir. RT-PCR ile gen ifade düzeyine bakıldı. Hücre süspansiyonunda biyokimyasal parametreler; glutatyon peroksidaz (GPx), katalaz (CAT), superoksit dismutaz (SOD), malondialdehit (MDA), nitrik oksit (NO), Miyelin peroksidaz (MPO) ve Prolidaz aktiviteleri çalışıldı. Hücre hatlarında apoptozis düzeyleri belirlekmek için Annexin V ve 7AAD içeren kiti, hücre döngüsü için BD Cycletest™ Plus DNA Reagent kiti kullanarak flov sitometride analiz edildi.Gen ifadesi analiz edildiğinde, MALAT1 ve B-MYB farklı kanser türlerinde farklı seviyede eksprese oldu. Biyokimyasal incelemelerin sonucunda; kanser ve kontrol hücre hatları arasında tüm parametreler açısından istatistiksel olarak anlamlı farklılıklar saptandı (p<0,05). Apoptozis ve hücre döngüsü sevilerinde bakıldığında, akciğer kanser hücre hattında apoptozis artmış proliferasyon azalmış, meme kanseri hücre hattında apoptozis değişmemiş, proliferasyon artmıştır. Kemik kanseri hücre hattında ise kanserli hücreler hem apoptozise gitmiş hemde proliferasyon yapmıştır.Sonuç olarak MALAT1 farklı kanserlerde farklı roller üstlenmektedir. MALAT1 akciğer kanserinde güçlü bir biyobelirteç olarak kullanabilirken, meme kanserinde proliferasyon için bir biyobelirteç, kemik kanserinde apoptozis için bir biyobelirteç olarak kullanabileceğini düşündürmektedir.
