IDENTIFICATION OF UPSTREAM CONSERVED NONCODING SEQUENCESFOR THE ANALYSIS OF TRANSCRIPTIONAL ACTIVATION ONIL-7 RECEPTOR ALPHA GENEH. brahim AksoylarInterleukin-7 (IL-7) is a key cytokine in the development of B- and T-lymphocytes. Responsiveness of lymphocyte populations to IL-7 is controlled by thetemporal and cell specific expression of the IL-7 receptor (IL-7R). Although there arestudies on the transcriptional regulation of IL-7R expression, these findings areinsufficient to explain the tight regulation on IL-7R expression.In this study, we identified conserved upstream sequences at the mouse IL-7Ralpha locus. We aligned murine and human genomic sequences using a pairwise globalalignment approach and found seven noncoding genomic regions with more than 85percent conservation. We amplified the conserved sequences from a specific bacterialartificial chromosome and constructed luciferase reporter vectors representing thesemouse genomic sequences. Here we investigate a reporter gene strategy to identify thetranscriptional activation properties of these mouse sequences in a mouse T-lymphomacell line.We evaluated the efficiency of luciferase assay system in a human kidneyfibroblast cell line and a mouse CD4 single positive T-lymphocyte cell line. Due to thedistinct expression properties of IL-7R, the present study can be expanded for cellsrepresenting the different stages of B- and T-lymphocyte development. Analysis of thetranscriptional responses of different cell lineages to the conserved sequences willfacilitate the studies on the investigation of the tight regulation on IL-7R expression.

IL-7R geninin transkripsyonel kontrolüile ilgili bir takım çalışmalar yapılmış olsa da, edinilen bulgular bu geninin üzerindekisıkı kontrolü açıklamak için yeterli değildir.Bu çalışmada, fare IL-7R alfa lokusunda, transkripsyon başlangıç noktasındanönde bulunan, korunmuş diziler aranmıştır. Fare ve insan genomunun bu bölgedekidizileri ikili genel hizalama yöntemine göre karşılaştırılmış olup, yedi tane yüzde 85'inüzerinde korunmuş kodlamayan dizi bulunmuştur. Bu diziler polimeraz zincirreaksyonu ile, spesifik bakteriyel yapay kromozomundan çoğaltılıp lusiferaz haberciplazmidlerine klonlanmıştır. Klonlanan korunmuş dizilerin fare T-lenfomahücrelerindeki transkripsyonel aktivasyon özelliklerini araştırmak için, lusiferaz geninedayanan bir genetik haberci stratejisi tasarlanmıştır.Belirlenen lusiferaz yöntemi, insan böbrek fibroblast hücrelerinde ve fare CD4pozitif T-lenfoma hücrelerinde denenmiştir. Bu çalışma, IL-7R geninin farklıekspresyon özelliklerine dayanarak genişletilmelidir. B- ve T-lenfosit gelişiminin farklıevrelerindeki hücrelerin korunmuş dizilere olan transkripsyonel duyarlılıklarınınincelenmesi, IL-7R geninin ekspresyonu üzerindeki kontrolü araştıran çalışmalarıhızlandıracaktır.
