Matrix metalloproteinases (MMPs) are zinc-bound proteinases that candisrupt the components of the extracellular matrix and numerous non-matrixproteins. Matrix Metalloproteinase-1 (MMP1) can directly break down fibrilcollagen and a few basic ECM components such as elastin, fibronectin, aggrecanand versikan. The polymorphism, known as G/2G at the position of the MMP1promoter region -1607, is affected by the expression or deletion of a guanine. Thepolymorphism known as 5A/6A in the MMP3 promoter region -1171 (-1612)originates from the deletion of an adenine base. Polymorphism, known as C / T inthe MMP9 promoter region at position -1562, takes place by replacing the Cnucleotide with the T nucleotide. Sleep Apnea Syndrome is a primary sleepdisorder characterized by respiratory pauses during sleep. Obstructive sleep apnea(OSA), the subtype of Sleep Apnea Syndrome, is an increasingly common form ofsleep-related respiratory distress. The mechanism that causes the Sleep ApneaSyndrome is not known exactly.In this study, we investigated whether promoter polymorphisms that affectthe gene expression level of MMP-1, MMP-3 and MMP-9 are related to sleepapnea syndrome. MMP-1 (-1607) G / 2G, MMP-3 (-1171) 5A/6A and MMP-9 (-1562) PCR-RFLP method was used to determine the genotype of the polymorphicregion in the promoter region of the C/T genes. No significant difference wasfound between the patient and control groups in terms of age and gender. BodyMass Index was found to be significantly higher in the patient group (p=0,01**).MMP1 -1607 G/G variant in genotype distribution was seen in 43.5% of patients.In the control group, 52.6% of the G / 2G variant was observed. Variantdistribution was statistically significant between the patient and control groups (p= 0.037). In the MMP3 -1171 genotype distribution, the 5A/6A variant was seenas 88.7% in the control group, but the variance distribution among the groups wasnot significant. MMP9 -1562 genotype distribution was observed more frequentlyin both patient and control groups. There was no statistically significant differencebetween the groups.

Uyku Apne Sendromu'nun alt tipi olan obstrüktif uyku apnesi (OSA) gittikçe yaygınlaşan uyku ile ilgili solunum bozukluğudur. Henüz Uyku Apne Sendromu'na neden olan mekanizma kesin olarak bilinmemektedir. Bu çalışmada MMP-1, MMP-3 ve MMP-9'ün gen ekspresyon seviyesini etkileyen promotor polimorfizmlerinin uyku apne sendromu ile ilişkisi araştırıldı.MMP-1 (-1607) G/2G, MMP-3 (-1171) 5A/6A ve MMP-9 (-1562) C/T genlerinin promotor bölgesindeki polimorfik bölgenin genotipini belirlemek için PZR-RFLP yöntemi kullanıldı. Hasta ve kontrol grubu yaş ve cinsiyet açısından değerlendirildiğinde anlamlı farklılık bulunmadı. Vücut Kitle İndeksi hasta grubunda anlamlı yüksek bulunmuştur (p=0,01**). MMP1 -1607 Genotip dağılımında G/G varyantı hastaların %43,5'inde görülmüştür. Kontrol grubunda ise G/2G varyantı %52,6'sında gözlenmiştir. Hasta ve kontrol grubu arasında varyant dağılımı istatistiksel olarak anlamlıdır (p=0,037). MMP3 -1171 genotip dağılımında 5A/6A varyantı kontrol grubunda %88,7 oranında görülmüş fakat gruplar arası varyant dağılımı anlamlı çıkmamıştır. MMP9 -1562 genotip dağılımında hem hasta hem de kontrol grubunda C/C varyantı daha sık izlendi. Gruplar arası varyant dağılımda istatistiksel olarak anlamlılık yoktu.
