Abstract

In this study native entomopathogenic nematodes (EPNs); Steinernema affine 46, S. feltiae 96, S. carpocapsae 1133 and Heterorhabditis bacteriophora 1144, were tested as plate and pots in climate chamber to determine their efficacy on the last instar larvae of the olive leaf moth, Palpita unionalis Hübner (Lepidoptera: Pyralidae) under laboratory conditions. Palpita unionalis larvae were collected from olive orchards in Çanakkale province. In laboratory conditions efficacy assays were done by using two application doses; 25 and 50 IJs/larva at 25 oC. In pot experiments ten P. unionalis larvae were used for each pot by dose of 40.000 IJs/pot at 25 oC.Mortality of infected P. unionalis larvae by entomopathogenic nematodes vary depending on nematode species and dose. In plate experiments; mortalities occurred on P. unionalis larvae caused by S. affine 46, S. carpocapsae 1133 and H. bacteriophora 1144 were found 91,6% and S. feltiae were 83,3%, by dose of 25 IJs and 100% by dose of 50 IJs for first three species and S. feltiae were 91,6%. In pot experiments; mortalities were determined as 50, 60, 100, 90%, respectively. Results of this study show that EPNs caused high mortality both in plate and pot experiments so it will be useful to determine the usage potential of these EPN species as biological control agent of P. unionalis by further studies that conducted in the field.

Bu çalışmada yerel entomopatojen nematod izolatları olan Steinernema affine 46 (Bovien, 1937), S. feltiae 96 (Filipjev, 1934), S. carpocapsae 1133 (Weiser, 1955) ve Heterorhabditis bacteriophora 1144 (Poinar, 1976) türlerinin zeytin fidan tırtılı Palpita unionalis (Hübner) son dönem larvaları üzerindeki etkinlikleri laboratuvar ve iklim odasındaki saksılarda araştırılmıştır. Palpita unionalis larvaları Çanakkale İli'ndeki zeytin bahçelerinden toplanmıştır. Laboratuvar koşullarında EPN etkinlikleri 12'li well platelerde 2 farklı uygulama dozu (25 IJ/larva ve 50 IJ/larva) ve 25 oC'de belirlenmiştir. Saksı denemesi ise her bir saksıya 10'ar adet P. unionalis larvası gelecek şekilde, tek uygulama dozu (40.000 IJ/saksı) ve 25 oC'de kurulmuştur.Entomopatojen nematodlar tarafından infekte edilen son dönem P. unionalis larvalarındaki ölüm oranları nematod türüne ve uygulama yoğunluğuna bağlı olarak değişmekle birlikte 25 IJ'de, S. affine, S. carpocapsae ve H. bacteriophora için %91,6, S. feltiae için 83,3 olarak tespit edilmiştir. Bu oran 50 IJ'de ilk üç izolat için %100, S. feltiae için 91,6 olarak bulunmuştur. Saksı denemesindeki ölüm oranları ise sırası ile %50, 60, 100 ve 90 olarak gözlemlenmiştir. Bu sonuçlar hem laboratuvar hem de saksı denemesinde EPN'lerin zararlıyı infekte etme oranının çok yüksek seviyelere ulaştığını ortaya koymuştur. Yapılacak doğa çalışmaları ile bu EPN türlerinin zararlının biyolojik mücadelesi kapsamında kullanım potansiyellerinin belirlenmesinde faydalı olacaktır.
