Abstract

Tomato is one of the most important product in terms of area, production and trade of fresh vegetable in Turkey. During the tomato production, several fungal and viral pathogens cause diseases, additionally, bacterial diseases result significant crop losses. One of the bacterial diseases is bacterial wilt and canker disease caused by Clavibacter michiganensis subsp. michiganensis (Smith) Davis et al., (Cmm). The resistant tomato plants are able to use for controlling the bacterial canker disease because the method is the most effective, economical, environmental, and human frendly. There is no disease-resistant tomato cultivars; however, resistant plants are present in wild tomato accessions. In this study, we used original tomato cultivar NCEBR3, mutant M3 and wild LA 1579 tomato lines to identify resistance reactions to the most virulent isolate (Cmm2) of bacterial canker disease in pathogenicity tests. The pathogenicity test results revaled that resistant M3 (?) plants and intermediate resistant LA1579 (?) plants where the two lines were crossed to produce F1 populations. Moleculer studies conducted to determine polimorphism between the parents, 11 Simple Sequence Repeats (SSR) markers were analysed in PCR reactions. The analyses shown that 8 SSR markers were polymorphic between NCEBR3, M3 and LA 1579 plants. Among tested markers, 73% of them gave polymorphism to use in future studies.

Domates, toplam ekiliş alanı, üretimi ve ticareti açıdan Türkiye'de yaş sebze grubunun en önemli ürünlerinden birisini oluşturmaktadır. Domateslerde yetiştiricilik sırasında fungal ve viral hastalık etmenlerinin yanı sıra pek çok bakteriyel etmende önemli ürün kayıplarına neden olmaktadır. Bu bakteriyel hastalıklardan biri olan Clavibacter michiganensis subsp. michiganensis (Smith) Davis et al.,'in (Cmm) neden olduğu bakteriyel kanser hastalığıdır. Bu hastalığın mücadelesinde dayanıklı çeşit kullanımı en etkin, ekonomik, çevre dostu ve insan sağlığını koruyan bir yöntem olarak görülmektedir. Bu hastalığa dayanıklı kültür domates çeşitleri bulunmamakta olup, bakteriyel kanser hastalığına karşı dayanıklılık kaynağı domatesin yabani akrabalarıdır. Çalışmada kültür domatesinin NCEBR3 homozigot hattı, bu domatesin kimyasal mutasyonu ile oluşturalan M3 mutant domatesleri ve yabani domates LA 1579 hattının Cmm' nin en virulent ırkı olan Cmm2'ye karşı patojenisite testlerinde reaksiyonları belirlenmiştir. Testler sonucunda dayanıklı M3 (?) hattı ile orta derecede dayanıklı LA 1579 (?) hattı melezlenerek F1 populasyonu oluşturulmuştur. Yapılan moleküler çalışmalarda ebeveynler arasında polimorfizmi ortaya koyabilmek için 11 adet Simple Sequence Repeats (SSR) markörü kullanılmış olup 8 adet SSR markörü NCEBR3, M3 ve LA 1579 arasında polimorfik bulunmuştur. Test edilen markörlerin ebeveynler üzerinde polimorfizm oranı %73 olarak ortaya konmuştur.
