Lung cancer has the leading mortality rate among all cancers and it is the second most common cause of death following deaths from cardiovascular diseases.This study was aimed to determine EGFR and HER2/neu oncogenes amplification that is significantly involved in carcinogenesis in several cancer types including lung cancers by using quantitative Real-Time PCR technique and probes and primers which are designed specifically for this genes.This study was performed by analyzing 100 archival samples which had been previously examined histopathologically and diagnosed as ?lung cancer? in İstanbul Yedikule Göğüs Hastalıkları Hastanesi. DNA extracts from 5 micron paraffin-embedded tissue sections which contain cancerous tissues were used as sample group.In the study, HER2/neu and EGFR gene amplification measured quantitatively and he amplification ratios were found % 11 and % 26, respectively. These findings obtained in the study is compatible with the literature.As a conclusion, it was determined that quantitative Real-Time PCR is a technique which can give fast, cheap, reliable and quantitative results for gene amplifications. This technique is an alternative technique for gene amplification determination among other techniques used for this purpose.

Akciğer kanseri, tüm dünyada mortalitesi en yüksek kanser türüdür ve kardiyovasküler hastalıklardan sonra ölüm nedenleri arasında ikinci sırada yer almaktadır.Çalışmanın amacı, kantitatif Real-Time PCR yöntemiyle akciğer kanserinin de içinde yer aldığı çok sayıda tipte karsinogenezis sürecinde önemli bir yere sahip olan EGFR ve HER2/neu onkogenlerine spesifik olarak dizayn edilmiş prob ve primerleriyle bu gen bölgelerinin amplifikasyonlarının incelenerek ortaya konmasıdır.Çalışmaya İstanbul Yedikule Göğüs Hastalıkları Hastanesinde, histopatolojik olarak incelenerek `akciğer kanseri' tanısı almış 100 hasta dahil edilmiştir. Her hastaya ait parafine gömülü doku örneklerinden, kanser dokusu içeren 5 mikronluk kesitlerden elde edilen DNA örnekleri çalışmaya alınmıştır.Çalışmada, Real-Time PCR ile kantitatif ölçümleri sonucu, akciğer kanserli bu dokuların % 18' inde HER2/neu gen amplifikasyonu, % 26' sında EGFR gen amplifikasyonları belirlenmiştir. Yapılan literatür taramasında da elde edilen bu sonuçlar literatür verileriyle çoğunlukla uyumlu bulunmuştur.Sonuç olarak kantitatif Real-Time PCR yöntemi, gen amplifikasyonlarının belirlenmesi için kullanılabilecek hızlı, güvenilebilir, ucuz ve nicel sonuç verebilecek bir yöntemdir. Elde edilen tüm verilere göre bu yöntem, gen amplifikasyonlarının belirlenmesine yönelik diğer yöntemler için, bu amaca yönelik alternatif bir yöntemdir.
