 أجرى هذا البحث لدراسة سلوك 8 تراكيب وراثية من قمح الخبز تحت الظروف البيئية لكل من رأس سدر و منطقة أشمون "بوسط الدلتا" خـلال موسمي 2014/2015 و 2015/2016 والتى تمت زراعتها باستخدام تصميم القطاعات الكاملة العشوائية لكل تجربة فى ثلاث مكررات , هذه التراكيب هي (مصر 1 ومصر 2 وشندويل 1 ونسر وسيوة 5 وسيوة 25 وS8/17 والسلالة 606). اوضحت النتائج ان هناك فروق معنوية بين التراكيب الوراثيةلجميع الصفات تحت الدراسة كما سجل معامل الاختلاف (CV) اعلى القيم لصفة المحصول البيولوجي يتبعه صفة محصول حبوب/النبات بينما كانت اقل القيم لصفة عدد الحبوب/نبات.اعطت السلالة 606 و S8/17 اعلى القيم لصفة محصول حبوب/النبات في كلا الموقعين ولا توجد فروق معنوية بينهما بينما اعطى الصنف مصر2 اقل القيم لنفس الصفة في كلا الموقعين. وبالنسبة لصفة المحصول البيولوجي/النبات سلكت التراكيب الوراثية نفس الاتجاه. وبالنسبة لصفة محصول الحبوب/النبات كانت هناك فروق معنوية عالية بين التراكيب الوراثية المدروسة في كلا الموقعين. و اعتمادا على دليل تحمل الإجهاد بالإضافة إلى قيمة محصول النبات بالرغم من ان التركب الوراثي مصر 1 كان ترتيبه الرابع تحت ظروف البيئة المثلى الا ان ترتيبه كان السادس تحت ظروف الاجهاد الملحي. و كذلك اعطي التركيب الوراثي مصر2 اعلى القيم لدليل الحساسية للاجهاد الملحىSSI) ) بينما اعطي اقل القيم لكل من متوسط الانتاجية (MP) والمتوسط الهندسي للانتاجية (GMP) و المتوسط المتوازن (HM)و دليل تحمل الاجهاد (STI) ودليل تحمل الاجهاد المعدل منسوبا الى البيئة المثلى (k1STI) ودليل تحمل االاجهاد المعدل منسوبا الى بيئة الاجهاد( k2STI) ودليل المحصول (YI) ودليل ثبات المحصول(YSI).ولهذا يمكن اعتبار ان مصر2 اكثر التراكيب الوراثية حساسية للاجهاد الملحي وعلى الجانب الاخر اعطت التراكيب الوراثية سيوه-25يتبعه سيوه-1اقل القيم لدليل الحساسية للاجهاد (SSI) بينما كان ترتيب التركيب الوراثيS8/17 الثاني بالنسبة للكفاءة المحصولية بعد السلالة -606 بدون فرق معنوي في كلا البيئتين وكان اعلى القيم في دليل ثبات المحصول (YSI). وشارك السلالة 606 القيم الاعلى في (MP, GMP, HM, STI, K1STI, K2STI, YI) ولهذا يمكن اعتبار التركيب الوراثي S8/17اعلى التراكيب تحملا للاجهاد الملحي. ولهذ تم اختيار خمسة تراكيب وراثية متباينة في تحملها للاجهاد الملحي السائد بمنطقة رأس سدر لدراسة التفريد الجزئي باستخدام IRAP. بتصنيف التراكيب الوراثية للقمح بإستخدام كاشفات التضخيم المتعدد الاشكال البينى للتروترانسبوزونات (IRAP) المعتمد على تكنيك تفاعل الاشكال المتعددة التسلسلى (PCR). تم فحص الحمض النووى الديوكسى ريبوزى (DNA) للتراكيب الوراثية الخمسة باستخدام ال PCR و خمسة بادئات لتكنيك ال (IRAP). وجد ان استخدام الخمس بادئات ادى الى انتاج 82 مكبرة (حزمة عديدة الاشكال) منها 36 مكبرة احادية الشكل. و يخنلف عدد الحزم المكبرة لكل بادئ من 13 لكل من البادئات (5-LTR2 and NIKITA) الى22 للبادئSUKULA . ووجد ان اعلى نسبه مئوية للتعدد المظهرى 80% للبادئ LTR6150. بينما اقل نسبه 30.8% سجلت للبادئ 5-LTR2. كما اوضحت النتائج ان الخمس بادئات IRAP انتجت 30 مكبرة او حزمة فريدة من الاشكال المتعددة لكل التراكيب الوراثية المستخدمة و ميزت ب 21 حزمة موجبة. و قد ميز البادئ LTR6150أكثر التراكيب الوراثية تحملا للملوحة سخا8/17 بواسطة خمسة حزم (مكبرة) الموجبة الفريدة AF1,2,3,10 and 12)). و بالنسبه لنسبة التماثل و التشابه الوراثى بين تراكيب القمح المستخدم وجد ان اقصى قيمة للتشابه 75.5% بين شندويل 1 ونسر, بينما اقل قيمة للتشابه الوراثى 52.1% لوحظت بين التركيبين الوراثيين مصر 2 و شندويل 1. قسم التحليل العنقودى بطريقة المجموعات الثنائية للمتوسط الحسابى (UPGMA) الى مجموعتان رئيسيتان واحدة منهم فصلت سخا8/17 كتركيب وراثى غير متشابه مع التراكيب الاخرى، بينما تفرقت المجموعه الثانية الى مجموعتان تحتية ، اشتملت الاولى على التركيب الوراثى مصر 1، نسر و شندويل وتحت المجموعة الثانية شملت مصر 2 و الذى يعتبر الثانى فى عدم التشابه مع التراكيب الاخرى. اثبتت قوة التمييز للIRAP انه من اكفأ التكنيكات للتمييز بين التراكيب الوراثية و ان البادئ LTR6150 اكفأ بادئ. و عليه يتضح أن كاشفات الIRAP كفء و ناجحة لدراسة التنوع و العلاقات الوراثية و انتاح البصمة الوراثية. و من الممكن استخدام النتائج المتحصل عليها فى اختيار الاباء المتميزة لتنفيذ برنامج تربية ذو فاعلية كبيرة لنبات القمح تحت الظروف الملحية .

 Two field experiments were conducted at the targeted two locations. The genotypes of bread wheat used in this study were (Misr-1, Misr-2, Shandweel-1, Nesr, Siwa-5, Siwa-25, S8/17 and Line – 606). Differentiation between eight bread wheat genotypes based on their yielding ability, salinity tolerance indices and molecular fingerprints were studied. Highly significant genotypic differences among the 8 genotypes for various characters were found. The highest coefficient of variation (CV) was shown by biological yield followed by grain yield/plant. With regard to grain yield per plant (g), under locations, S8/17 and Line – 606 genotypes gave the highest yield with insignificant difference, while Misr-2 was the lowest one under adequate and stress affected conditions. The genotypic performance had the same trend for biological yield (g)/plant under the salt stress conditions prevailing at Ras-Sudr. Based on the stress tolerance index (STI) and grain yield although Misr-1 genotype exhibited the fourth rank under adequate location, it ranked the sixth under salt stress affected location. This was confirmed by detecting the highest value of Stress susceptibility index (SSI) with Misr-2and the lowest values of eight stress tolerance indices. Therefore, it could be considered as the most salt stress sensitive genotype. On the contrary, Siwa-25 followed by Siwa-1 genotypes achieved the lowest values of SSI index. While, S8/17 genotype ranked second for mean yield performance under both environments and had the highest values of YSI and shared with L-606 as the highest ones in MP, GMP, HM, STI, K1STI, K2STI and YI stress tolerance indices, S8/17 genotype could be considered as the highest salt tolerant one. So, we have selected five genotypes with contrasting performance under prevailing salt stress at Ras Sudr conditions for molecular analysis by IRAP markers. It was essential to characterize the five bread wheat genotypes using PCR-based markers such as IRAPs. A total of 82 amplicons (amplified fragments) were generated via the five primers, out of them 36 have been monomorphic. The number of amplicons per primer varied from 13 for each of primers (5-LTR2 or NIKITA) to 22 for primer (SUKULA). The highest percentage of polymorphism (80%) was detected with the primer (LTR 6150), while the lowest one (30.8%) was recorded in the primer (5-LTR2). Also, these results show that IRAP profiles obtained with the five different long terminal repeats (LTR) primers yielded 30 unique bands out of the total amplified fragments among all the genotypes and distinguished by (21 M+). The primer (LTR6150) discriminated S8/17 (most tolerant genotype) by 5 unique bands of amplicon fragments (AF1, 2,3,10 and 12). The percentage of similarity between the studied genotypes revealed that, the maximum value of similarity was 75.8% observed between Shandaweel-1 and Nesr, whereas the minimum value was 52.1% observed between Misr-2 and Shandaweel-1 genotypes. The UPGMA dendrogram classified the five bread wheat genotypes into two main clusters. One of them separated S8/17 as a genetically dissimilar genotype with all other ones. While the second cluster was separated into two sub-clusters, comprised the first involved Misr-1, Nesr and Shandaweel-1 genotypes and the second sub-cluster separated Misr-2 which was described as the second dissimilar one with all other genotypes tested. The calculated IRAP discrimination power was more efficient and the most efficient primer was LTR6150. It is clearly concluded that IRAP markers have been used successfully for assessing genetic diversity and relationships among bread wheat genotypes. Also, we could use these results for selecting the elite parents to generate an effective breeding wheat program under saline conditions.
